Comments: "Preparation of monoclonal antibody. Lagenar maculae were dissected from the inner ears of 1- to 3-d-old posthatch chicks in PBS (150 mm NaCl, 10 mm sodium phosphate, pH 7.2) containing 2 mm benzamidine, 10 μmpepstatin, 1 μg/ml leupeptin, and 0.1 mmphenylmethylsulfonylfluoride, stored under liquid N2 until required, and used to prepare a crude membrane fraction by differential centrifugation. The membrane fraction was suspended in PBS and used to immunize a BALB/c mouse. The mouse was injected three times at 1 month intervals using the material from ∼250 lagenar maculae for each injection. The mouse was then rested for a period of 5 months, boosted with a membrane fraction prepared from ∼350 lagenar maculae, and 7 d later the spleen was used for the preparation of hybridoma cells using standard techniques (Kohler and Milstein, 1975)."
