Sample type |
RNA |
Source name |
mouse primary liver cells
Organism |
Mus musculus |
Characteristics |
treatment: 4.5% EtOH, 5Hr, fresh medium 6Hr
Treatment protocol |
Mouse primary liver cells were treated with 4.5% ethanol for 5 hours (R0) and then washed and cultured in fresh DMEM F12 10% FBS medium for 3 (R3), 6 (R6), 24 (R24), and 48 (R48) hours.
Growth protocol |
Primary liver cells were isolated from BALB/c mice by collagenase B according to the manufacturer’s instructions (Worthington Biochemical, Lakewood, NJ) and cultured as described (PMID: 22535522, DOI: 10.1091/mbc.E11-11-0926)
Extracted molecule |
total RNA |
Extraction protocol |
RNA of all samples was extracted with Trizol reagent. Total RNA was isolated and purified by RNeasy Mini Kit (Qiagen, Cologne, Germany).
Label |
Cy3
Label protocol |
The RNA was labelled with the SABiosciences C-03 RT² First Strand Kit (SABiosciences - QIAGEN, Frederick, MD) standard protocol
Hybridization protocol |
Labeled cRNA was combined with hybridization buffer and hybridized to the array at 58°C for 16-20 hours with rocking
Scan protocol |
Arrays were scanned with the Illumina iScan BeadArray scanner standard protocol
Data processing |
Data were extracted with the Illumina BeadStudio with Gene Expression module, average normalization, to create an output file. These values were imported into Spotfire DecisionSite v9.1.2 and log2 transformed.
