Design: Library preparation and sequencingONT-RegularA total amount of 3-4 g DNA per sample was used as input material for the ONT library preparations. After the sample was qualified, size-select of long DNA fragments were performed using the PippinHT system (Sage Science, USA). Next, the ends of DNA fragments were repaired, and A-ligation reaction were conducted with NEBNext Ultra II End Repair/dA-tailing Kit (Cat# E7546). The adapter in the SQK-LSK109 (Oxford Nanopore Technologies, UK) was used for further ligation reaction and DNA liabrary was measured by Qubit 4.0 Fluorometer (Invitrogen, USA). About 700ng DNA library was constructed and performed on a Nanopore PromethION sequencer instrument (Oxford Nanopore Technologies, UK) at the Genome Center of Grandomics (Wuhan, China).

Study: Chromosome-level genome assembly of Chouioia cunea Yang, the parasitic wasp of the fall webworm

Chouioia cunea Yang 1989 is a parasitic wasp and natural enemy of several lepidopteran pests during their pupal stage, this wasp has successfully used to control pests such as the fall webworm. Here we reported the first chromosome-level genome of Eulophidae. We applied short, long, chromatin-linked sequencing reads and transcriptomic data to assemble the chromosome-level genome of C. cunea. The version of the genome assembly of C. cunea was highly contiguous with a total assembly length of 171.99 Mb contained in 11 contigs with a GC content of 36.89%, the scaffold/contig N50 length was 31.70/26.52Mb. The Benchmarking Universal Single-Copy Orthologs completeness score is 98.70% at the protein level, including 1,318 single-copy genes and 32 duplicated genes. Repeat sequences accounted for 15.15% of the whole-genome assembly. A total of 12,258 protein-coding genes were annotated in this genome. We also predicted 324 noncoding RNAs including 84 micro-RNAs, 69 ribosomal RNAs, 28 small nuclear RNAs, 205 transfer RNAs, and 44 others. This high-quality genome of C. cunea is an important step toward bettering our knowledge of the Eulophidae, it will serve as a valuable resource for phylogenetic relationships and a better understanding of the evolution of Chalcidoidea.
