SRX4717057: RNA-Seq of human Vdelta1 T cells from peripheral blood. Donor 6, delta chain

Design: Flow cytometric sorted cells were subjected to RNA extraction using the RNAeasy mini kit (Qiagen). Next, 12l mRNA of each sample was reverse transcribed with the Superscript III enzyme kit (Invitrogen). CDR3 TRG and TRD sequencing amplicons were generated as described previously (Ravens et al. Nat Immunology 2017), while using 25 30 PCR cycles. According to Illumina guidelines 96-samples were labeled with Nextera XT indices and subjected to Illumina MiSeq analysis (500 cycle; paired-end sequencing), while 20% PhIX was added as a control.
